[Abstract]

Background and ObjectiveEndometrial cancer (EC) is one of the most frequent gynecologic malignancies in affluent communities. Endometrial carcinoma, cervical cancer and ovarian tumor are the three most malignant tumors in female reproductive duct. It is 20%-30% of female procreant system malignant tumor in china. Most cancers occur in pre-and post-menopausal women and become a major disease that treaten old female lives. From 1960 year, the outbreak rate was reported the ascending trend. Its incidence in many countries began to exceed that attributable to cervical cancer. Up to date, the certain cause of EC is still unknow. Epidemiological survey and experiment indicate that hormonal factors leading role in initiation /promotion of EC belongs to the excessive estrogenic stimulation. Hormone synthesis and metabolism are very complex. They need many enzyme in the process. The gene polymorphism can influnce the activity of enzyme and contribute to endometrial cancer susceptibility.We chose the candidate genes associated with androgen synthesis CYP17(17?hydroxykase-17,20 lyase gene) and CYP19(CYP450 gene). The purpose of this study is to investigate the relationsip between the two genes and EC and to provide theoretic base to prevent and treat EC. Materials and methodsOne hundred and seven cases of normal women, one hundred and four cases of EC were enrolled from the second affiliated hospital of Zhengzhou University and the first affiliated hospital of Zhengzhou University ,who were patients from the January of 2004 to the june of 2006. On the second morning 2ml intravenous blood was extracted, anti-solidated with EDTA-K2, stocked in the -80?refrigerate. All subjects were the Han nationality without hormone replacement therapy, did not have tumor, sever cute or chronic disease. The women with endometrial cancer did not have radiation, chemotherapy or hormone therapy. The genomic DNA was isolated from peripheral blood collected above. Polymerase chain reaction and restriction fragment length polymorphism (PCR?RFLP) were employed to detect the mutation of CYP17 gene and CYP19 gene. Fragments of CYP17 gene and CYP19 gene were amplified by PC, then the PCR product of CYP17 gene digested with restriction endonuclease MspA1I. Finally the digestion product was subjected to electrophoresis on a gel containing2% agarose. The PCR product of CYP 19 were separated in the 15% polyacrilamide (30:1) nondenaturing gel and visualized by ethidium bromide staining. tThen we confirmed the genotype. This approach allowed the separation of seven CYP19 alleles of different sizes starting from the shortest (A1, 123 bp) to the longest (A7, 146 bp) ones.
